Foundayo® is a once-daily prescription tablet containing orforglipron, a GLP-1 receptor agonist. It is licensed for weight loss and weight maintenance in adults with a BMI of 30 or above, or a BMI between 27 and 30 with at least one weight-related condition, alongside a reduced-calorie diet and increased physical activity. The same MHRA authorisation also covers improving blood sugar control in adults with insufficiently controlled type 2 diabetes.
GLP-1 receptor agonists mimic a hormone the body releases after eating. Orforglipron acts on the areas of the brain that regulate appetite, helping people feel fuller for longer, reducing hunger and decreasing food cravings — so they eat less alongside appropriate diet and lifestyle measures.
In the UK, Foundayo® is only available following a clinical assessment. It is not currently available on the NHS; that decision follows a separate evaluation by NICE.
Why Foundayo® Benefits you need to know aboutNo fasting window — take it whenever suits you
Foundayo® is the only GLP-1 pill that can be taken at any time of day, with or without food and with no water restrictions. Orforglipron is a small molecule rather than a peptide, so it does not need an absorption enhancer or an empty stomach — which is why it avoids the 30-minute morning wait required by oral semaglutide.
No needles — a daily tablet instead
For people who would rather not inject, Foundayo® offers GLP-1 treatment in a once-daily oral form. No pens, no needles, no sharps disposal and no refrigerated delivery.
Weight loss proven in a large phase 3 programme
Across two 72-week registration trials enrolling more than 4,500 adults, orforglipron produced significantly greater weight loss than placebo at every dose studied — up to 25 lb, around 11 kg or 11.1% of body weight, in adults without diabetes on the 17.2 mg dose.
Improvements in other health measures
Adults taking orforglipron alongside diet and exercise also saw improvements in waist circumference, non-HDL cholesterol, triglycerides and systolic blood pressure. These findings are from clinical trial data; Foundayo® is not licensed to treat cholesterol or blood pressure.
Greater fat mass loss than lean mass loss
Trial data indicate the weight reduction seen with orforglipron involves greater loss of fat mass than lean mass. This result is based on a less rigorous assessment and may be due to chance.
Foundayo® is backed by the ATTAIN phase 3 programme. Here is what the evidence shows.
| 17.2 mg at 72 weeks | Placebo |
|---|---|
| 11.1% average weight loss — up to 25 lb (11 kg) | 2.1% average weight loss — 5.3 lb (2.4 kg) |
In ATTAIN-1, a 72-week trial in adults with obesity or overweight and a weight-related condition but without diabetes, average body weight loss was 7.4% on the 5.5 mg dose, 8.3% on 9 mg and 11.1% on 17.2 mg, compared with 2.1% on placebo. Based on an average starting weight of 227 lb (about 103 kg), that is up to 25 lb — around 11 kg, or roughly 1 st 11 lb — on the highest dose.
About 1 in 5 adults (18.4%) taking the 17.2 mg dose lost 20% or more of their body weight — roughly 45 lb — compared with 2.9% of those on placebo. Among the 1,127 participants who had prediabetes when they joined the study, up to 91% reached near-normal blood sugar levels by 72 weeks, versus 42% on placebo.
ATTAIN-1 randomised 3,127 adults across nine countries to one of three orforglipron doses or placebo, all alongside a reduced-calorie diet and increased physical activity. Participants had obesity, or overweight with at least one weight-related condition such as high blood pressure, high cholesterol, obstructive sleep apnoea or cardiovascular disease. Adults with diabetes were excluded.
ATTAIN-2 studied 1,613 adults with a BMI of 27 kg/m² or above and type 2 diabetes across 136 sites in ten countries. Average weight loss was 5.1%, 7.0% and 9.6% across the three doses versus 2.5% on placebo, and HbA1c fell by 1.3–1.8% from an average baseline of 8.1%.
